Garage 10 sits at 6700 MD Anderson Blvd, open 24 hours daily. It connects directly to MD Anderson Cancer Center by a crosswalk on the third floor, which makes it a strong choice if that's your destination specifically.
Garage 16 sits at 6701 Fannin St, underground beneath the Wallace Tower and Texas Children's Hospital. Being underground means direct indoor access without crossing a street at all, useful in Houston's summer heat or during sudden rain.

Both garages share the same posted rate structure. One hour costs $5. Two hours runs $9. Three hours is $10, and four hours is $12. Six hours costs $14, eight hours is $15, and ten hours runs $17. Twelve hours costs $18, and the full 24-hour maximum is $19.
That structure rewards a longer stay at both garages. The jump from one hour to two only adds $4. The jump from two hours to three only adds a dollar. Once you're past the first hour or two, the per-hour cost drops fast.
A lost ticket costs $25 at Garage 10. This is separate from a SmartChip system, so keep your physical ticket if that's what you're issued, since losing it costs real money on top of your parking time. A quick photo of the ticket with your phone is an easy backup in case the original goes missing.

Location should drive your choice between these two. If your destination is MD Anderson Cancer Center specifically, Garage 10's direct crosswalk access makes it the more convenient pick.
